PNC-27 Dosage Protocol
PNC-27 is a synthetic peptide combining the p53 MDM2-binding domain with a membrane-residence sequence, studied for selective induction of necrosis in cancer cells.
⚡ Quickstart Highlights
Dosing & Reconstitution Guide
Route: Subcutaneous | Frequency: Once daily | Half-life: Variable
Standard Approach (3 mL = 10.00 mg/mL)
Reconstituting with 3 mL bacteriostatic water produces a concentration of 10.00 mg/mL. Volume per dose changes with concentration; mg dose itself does not change between vial sizes.
| Phase / Protocol | Dose | U-100 Units | Volume | Doses per vial |
|---|---|---|---|---|
| Weeks 1–2 (titration) | 100 mcg | 1 units | 0.010 mL | 300 doses |
| Weeks 3–4 | 200 mcg | 2 units | 0.020 mL | 150 doses |
| Weeks 5–8 | 300 mcg | 3 units | 0.030 mL | 100 doses |
| Weeks 9–12 | 400 mcg | 4 units | 0.040 mL | 75 doses |
| Weeks 13–16 (target) | 500 mcg | 5 units | 0.050 mL | 60 doses |
Reconstitution Steps
- Wipe the vial stopper and BAC water vial with alcohol; let dry.
- Draw 3 mL of bacteriostatic water into a sterile syringe.
- Inject slowly down the inside glass wall of the peptide vial. Do not aim at the powder.
- Gently swirl until fully dissolved. Do not shake.
- Label with reconstitution date. Refrigerate at 2–8°C; use within 30 days.
Supplies Needed
Estimates for an 8-week and 12-week cycle at 300 mcg per dose, once daily (7 doses/week).
| Item | 8-Week Cycle | 12-Week Cycle |
|---|---|---|
| PNC-27 () vials | 1 vials | 1 vials |
| Insulin syringes (U-100) | 56 | 84 |
| Bacteriostatic water (10 mL) | 1 × 10 mL | 1 × 10 mL |
| Alcohol swabs | 1 × 100-pack | 2 × 100-pack |
Protocol Overview
PNC-27 is preclinical research only. No established human clinical protocol. Use is research-only.
Dosing Protocol
Research protocols vary by study design. Not for use outside research settings.
Storage Instructions
| State | Temperature | Duration |
|---|---|---|
| Lyophilized | −20°C (−4°F) | Up to 24 months, dry & dark |
| Reconstituted | 2–8°C (35–46°F) | Up to 30 days, protect from light |
Important Notes
How This Works
PNC-27 fuses the p53 N-terminal MDM2-binding region (residues 12-26) with the leader sequence of penetratin or other membrane-residence peptides. The result selectively binds HDM-2 expressed at the cancer-cell membrane (a tumor-specific phenomenon) and triggers necrosis without affecting normal cells lacking surface HDM-2.
Potential Benefits & Side Effects
Potential Benefits
- Cancer-cell-selective necrosis in preclinical models.
- Spares normal cells lacking surface HDM-2.
- Studied in multiple solid tumor models.
Side Effect Profile
- Limited human safety data.
- Investigational compound — research only.
Lifestyle Factors
- Use is research only; not a standalone treatment.
Injection Technique
- Research protocol dictates route.
