PE 22-28 (10 mg Vial) Dosage Protocol
PE 22-28 is a 7-amino-acid analog of spadin, studied as a fast-acting antidepressant via TREK-1 potassium channel inhibition.
⚡ Quickstart Highlights
Dosing & Reconstitution Guide
Route: Subcutaneous | Frequency: Daily during cycle | Half-life: ~6 hours
Standard Approach (3 mL = 3.33 mg/mL)
Reconstituting with 3 mL bacteriostatic water produces a concentration of 3.33 mg/mL. Volume per dose changes with concentration; mg dose itself does not change between vial sizes.
| Phase / Protocol | Dose | U-100 Units | Volume | Doses per vial |
|---|---|---|---|---|
| Weeks 1–2 (titration) | 50 mcg | 1.5 units | 0.015 mL | 200 doses |
| Weeks 3–8 (target) | 100 mcg | 3 units | 0.030 mL | 100 doses |
| Weeks 9–12 (optional) | 150 mcg | 4.5 units | 0.045 mL | 66 doses |
| Weeks 13–16 (optional) | 200 mcg | 6 units | 0.060 mL | 50 doses |
Reconstitution Steps
- Wipe the vial stopper and BAC water vial with alcohol; let dry.
- Draw 3 mL of bacteriostatic water into a sterile syringe.
- Inject slowly down the inside glass wall of the peptide vial. Do not aim at the powder.
- Gently swirl until fully dissolved. Do not shake.
- Label with reconstitution date. Refrigerate at 2–8°C; use within 14 days.
Supplies Needed
Estimates for an 8-week and 12-week cycle at 150 mcg per dose, daily during cycle (7 doses/week).
| Item | 8-Week Cycle | 12-Week Cycle |
|---|---|---|
| PE 22-28 (10 mg) vials | 1 vials | 2 vials |
| Insulin syringes (U-100) | 56 | 84 |
| Bacteriostatic water (10 mL) | 1 × 10 mL | 1 × 10 mL |
| Alcohol swabs | 1 × 100-pack | 2 × 100-pack |
Protocol Overview
PE 22-28 is preclinical-only at present — no human clinical trials. Animal data shows fast-onset antidepressant effects (days vs weeks for SSRIs) via TREK-1 channel blockade.
Research protocols use 2–4 week cycles given limited long-term data.
Dosing Protocol
Daily titration (subcutaneous):
- Weeks 1–2 (titration): 50 mcg/day.
- Weeks 3–8 (target): 100 mcg/day. Most-cited research dose.
- Weeks 9–12 (optional): 150 mcg/day.
- Weeks 13–16 (optional): 200 mcg/day. Upper end of researched range.
Cycle structure: 8–12 weeks active, then 4-week break for reassessment. Long-term continuous use not validated.
Storage Instructions
| State | Temperature | Duration |
|---|---|---|
| Lyophilized | −20°C (−4°F) | Up to 24 months, dry & dark |
| Reconstituted | 2–8°C (35–46°F) | Up to 14 days, protect from light |
Important Notes
How This Works
PE 22-28 is a heptapeptide derived from spadin, the C-terminal fragment of sortilin. It selectively blocks the TREK-1 (KCNK2) potassium channel, which is highly expressed on serotonergic neurons. TREK-1 blockade enhances serotonergic neurotransmission and produces antidepressant effects in animal models — with onset much faster than SSRIs.
Potential Benefits & Side Effects
Potential Benefits
- Fast-acting antidepressant in rodent models (effects in days vs weeks).
- TREK-1 selective — distinct mechanism from SSRIs.
- No sedation or weight effects in available research.
Side Effect Profile
- Generally well-tolerated in animal research.
- Limited human clinical data.
Lifestyle Factors
- Pair with established mental-health practices (sleep, exercise, social connection).
- Animal data; not yet established in human trials.
Injection Technique
- Subcutaneous, rotate sites.
