FOXO4-DRI (10 mg Vial) Dosage Protocol
FOXO4-DRI is a synthetic senolytic peptide that disrupts the FOXO4-p53 interaction, selectively triggering apoptosis in senescent cells while sparing healthy ones.
⚡ Quickstart Highlights
Dosing & Reconstitution Guide
Route: Subcutaneous | Frequency: Once daily | Half-life: ~8 hours
Standard Approach (3 mL = 3.33 mg/mL)
Reconstituting with 3 mL bacteriostatic water produces a concentration of 3.33 mg/mL. Volume per dose changes with concentration; mg dose itself does not change between vial sizes.
| Phase / Protocol | Dose | U-100 Units | Volume | Doses per vial |
|---|---|---|---|---|
| Weeks 1–4 (titration) | 250 mcg | 7.5 units | 0.075 mL | 40 doses |
| Weeks 5–8 | 375 mcg | 11.2 units | 0.11 mL | 26 doses |
| Weeks 9–16 (target) | 500 mcg | 15 units | 0.15 mL | 20 doses |
Reconstitution Steps
- Wipe the vial stopper and BAC water vial with alcohol; let dry.
- Draw 3 mL of bacteriostatic water into a sterile syringe.
- Inject slowly down the inside glass wall of the peptide vial. Do not aim at the powder.
- Gently swirl until fully dissolved. Do not shake.
- Label with reconstitution date. Refrigerate at 2–8°C; use within 30 days.
Supplies Needed
Estimates for an 8-week and 12-week cycle at 375 mcg per dose, once daily (7 doses/week).
| Item | 8-Week Cycle | 12-Week Cycle |
|---|---|---|
| FOXO4-DRI (10 mg) vials | 3 vials | 4 vials |
| Insulin syringes (U-100) | 56 | 84 |
| Bacteriostatic water (10 mL) | 1 × 10 mL | 2 × 10 mL |
| Alcohol swabs | 1 × 100-pack | 2 × 100-pack |
Protocol Overview
FOXO4-DRI uses cycled dosing — typically 3 doses per week for 3 weeks, then a long break. The mechanism (selective senescent-cell apoptosis) does not require continuous administration; once senescent cells are cleared, the body needs time to fully process the changes.
Limited human clinical data — most evidence is preclinical mouse studies.
Dosing Protocol
Daily titration (subcutaneous):
- Weeks 1–4 (titration): 250 mcg/day.
- Weeks 5–8: 375 mcg/day.
- Weeks 9–16 (target): 500 mcg/day. Upper end of preclinical-derived range.
Cycle structure: 12–16 weeks active, then 8–12 week break. Some users run 1–2 cycles per year.
Storage Instructions
| State | Temperature | Duration |
|---|---|---|
| Lyophilized | −20°C (−4°F) | Up to 24 months, dry & dark |
| Reconstituted | 2–8°C (35–46°F) | Up to 30 days, protect from light |
Important Notes
How This Works
FOXO4-DRI is a D-retro-inverso peptide that disrupts the FOXO4-p53 protein interaction. In senescent cells (which accumulate with age and contribute to tissue dysfunction), this interaction sequesters p53 and prevents apoptosis. Disrupting the interaction releases p53 to drive selective senescent-cell apoptosis.
Healthy cells lack the FOXO4-p53 dependency and are spared. Animal studies show improved organ function, hair regrowth, and physical performance after senolytic dosing in aged mice.
Potential Benefits & Side Effects
Potential Benefits
- Selective senescent-cell apoptosis.
- Improvements in physical performance in aged mice.
- Hair regrowth and improved kidney function in aged-mouse studies.
- Cycled dosing — does not require continuous administration.
Side Effect Profile
- Limited human safety data.
- Injection-site reactions possible.
- Theoretical concerns with rapid senescent-cell clearance in patients with high senescent burden.
Lifestyle Factors
- Cycled dosing (3×/week × 3 weeks, then break) is standard.
- Pair with anti-inflammatory and longevity-supportive lifestyle.
Injection Technique
- Subcutaneous, rotate sites.
