Adipotide (10 mg Vial) Dosage Protocol
Adipotide is a synthetic peptidomimetic that targets prohibitin on blood vessels within white adipose tissue, delivering a pro-apoptotic sequence that destroys those vessels. Preclinical studies showed ~30% body weight reduction in obese rodents and ~11% in obese primates. Human clinical development was discontinued due to reversible kidney toxicity at higher doses.

Dosing & Reconstitution Guide
Route: Subcutaneous | Frequency: Once daily | Half-life: Short
Standard Approach (3 mL = 3.33 mg/mL)
Reconstituting with 3 mL bacteriostatic water produces a concentration of 3.33 mg/mL. Volume per dose changes with concentration; mg dose itself does not change between vial sizes.
| Phase / Protocol | Dose | U-100 Units | Volume | Doses per vial |
|---|---|---|---|---|
| Weeks 1–2 (titration) | 250 mcg | 7.5 units | 0.075 mL | 40 doses |
| Weeks 3–4 | 500 mcg | 15 units | 0.15 mL | 20 doses |
| Weeks 5–6 | 750 mcg | 22.5 units | 0.22 mL | 13 doses |
| Weeks 7–8 (target max) | 1 mg | 30 units | 0.30 mL | 10 doses |
Reconstitution Steps
- Wipe the vial stopper and BAC water vial with alcohol; let dry.
- Draw 3 mL of bacteriostatic water into a sterile syringe.
- Inject slowly down the inside glass wall of the peptide vial. Do not aim at the powder.
- Gently swirl until fully dissolved. Do not shake.
- Label with reconstitution date. Refrigerate at 2–8°C; use within 30 days.
Supplies Needed
Estimates for an 8-week and 12-week cycle at 750 mcg per dose, once daily (7 doses/week).
| Item | 8-Week Cycle | 12-Week Cycle |
|---|---|---|
| Adipotide (10 mg) vials | 5 vials | 7 vials |
| Insulin syringes (U-100) | 56 | 84 |
| Bacteriostatic water (10 mL) | 2 × 10 mL | 3 × 10 mL |
| Alcohol swabs | 1 × 100-pack | 2 × 100-pack |
Protocol Overview
Adipotide is a peptidomimetic that targets prohibitin on adipose tissue vasculature, destroying blood vessels feeding white fat. Preclinical studies demonstrated dramatic weight loss (~30% in rodents, ~11% in primates) over short cycles.
Important safety note: Human clinical development was discontinued after Phase 1 due to reversible kidney toxicity at higher doses. This protocol represents an extremely cautious educational reference — not a validated human treatment.
Dosing Protocol
Cautious daily titration (subcutaneous, once daily):
- Weeks 1–2 (titration): 250 mcg/day. Start conservatively; monitor for renal signs.
- Weeks 3–4: 500 mcg/day if tolerated.
- Weeks 5–6: 750 mcg/day if tolerated.
- Weeks 7–8 (target max): 1000 mcg (1 mg) /day. Do not exceed.
Cycle: 4–8 weeks maximum. Extend only with extreme caution due to preclinical renal effects.
Timing: Once daily at any consistent time; rotate injection sites.
Monitoring: Consider periodic kidney function assessment for any cycle beyond 4 weeks.
Storage Instructions
| State | Temperature | Duration |
|---|---|---|
| Lyophilized | −20°C (−4°F) | Up to 24 months, dry & dark |
| Reconstituted | 2–8°C (35–46°F) | Up to 30 days, protect from light |
Important Notes
How This Works
Adipotide binds prohibitin-1 on the vascular endothelium of white adipose tissue and delivers a pro-apoptotic sequence that triggers vessel destruction. Fat cells lose their blood supply, undergo apoptosis, and are metabolized. Preclinical models show highly targeted effects sparing other tissues.
